Constriction of Human Umbilical Arteries

چکیده
Experiments on perfused segments or isolated rings of human umbilical artery showed that the magnitude of their contraction in response to bradykinin, epinephrine, or serotonin depended on the P02 of the perfusing or bathing solution. Constriction to bradykinin varied directly with the O2 tension and was greatest when the Po2 was approximately that of umbilical arterial blood during the transition from the fetal to the neonatal period. Catecholamine beta-receptors were not demonstrated (not present or not functional) in the umbilical artery, since there was no vascular response to either isoproterenol or to propranolol. Alpha-receptors were not required for O2 or bradykinin constriction since these agents constricted the umbilical artery after enough phentolamine had been given to block constriction by norepinephrine. Because of the interaction between Oo and vasoconstrictor agents the O2 environment of umbilical vessels must be rigidly controlled in experiments measuring vascular reactivity. Sensitization by O2 to the effects of chemical substances endogenous to the fetus or mother may play an important role in circulatory adjustments essential for neonatal life. ADDITIONAL

Noscapine antagonizes vasoconstrictor action of bradykinin in isolated human umbilical artery
Abstract Background: It has been demonstrated that noscapine, an antitussive opioid alkaloid, could antagonize bradykinin- induced responses such as bradykinin effects in guinea-pig ileum, cough induced by bradykinin receptor agonist and angiotensin converting enzyme inhibitors, and brain damage after brain edema both in neonatal rat model and in patients with stroke. In the present study, the...
متن کاملRelationship between erythropoietin and nitric oxide in the contraction of rat renal arcuate arteries and human umbilical vein endothelial cells.
We have investigated the effects of recombinant human erythropoietin (EPO) on the responses of rat renal arcuate arteries to dopamine, noradrenaline and acetylcholine and on the release of NO from human umbilical vein endothelial cells (HUVEC) in culture. Noradrenaline induced a concentration-dependent constriction and acetylcholine a concentration-dependent relaxation of the vessels. The effec...
متن کاملShengmai Suppressed Vascular Tension in Umbilical Arteries and Veins of Human and Sheep
Objective—The umbilical cord is a critical pathway between mothers and fetuses, and regulations of umbilical vessel tension are important for fetal growth. Shengmai is an herbal medicine being used in treatments of cardiovascular diseases. However, effects of Shengmai on human blood vessels and related pharmacological mechanisms are unclear. Methods—This study investigated the effects of relate...

The vasoactive effects of oxidative stress induced by hydrogen peroxide (H2O2) on human umbilical artery strips as well as the possible mechanisms involved are studied. Contraction responses to cumulative H2O2 (10 M 3 × 10 M) in endothelium intact and denuded umbilical arteries and responses to cumulative H2O2 after incubation with L-NAME (10 M) (n = 8), indomethacin (10 M) (n = 8) and verapami...
متن کاملGuanosine 3',5'-monophosphate and adenosine 3',5'-monophosphate content of human umbilical artery.
Human umbilical arteries are unique vessels in that they close quickly and completely at birth. It has been suggested that cyclic uanosine 3',5'-monophosphate (cAMP) in relaxation. This hypothesis has been evaluated in term gestational human umbilical artery segments incubated at 37 degrees C and in room air. (a) The basal cGMP content (1 pmol/mg protein) of artery segments incubated in room ai...
